The Toronto Blue Jays selected him with the 11th overall choice. On July 27, he made his professional debut with the Class A-Short Season Northwest League’s Vancouver Canadians after signing a $4.55 million contract. Manoah had a 2.65 ERA in six starts with 27 strikeouts in 17 innings. Due to the minor league season being postponed in 2020 as a result of the COVID-19 epidemic, Manoah did not participate in any games.

Manoah was given a job with the Triple-A East’s Buffalo Bisons to start the 2021 season. Over six scoreless innings in his first start of the year, he struck out 12 batters. Manoah had a 3-0 record, a 0.50 ERA, and 27 strikeouts over 18 innings in three starts for Buffalo.
